In 1904, de Diego was elected to the Puerto Rican Senate. He served as President of the Senate from 1909 to 1916. He was also a member of the Puerto Rican Academy of Languages.
De Diego was a supporter of Puerto Rican autonomy. He helped draft the Puerto Rican Constitution of 1917. He also advocated for Puerto Rican citizenship and voting rights.
